Chemokines in the cerebrospinal fluid of patients with active and stable relapsing-remitting multiple sclerosis BJMBR
Moreira,M.A.; Souza,A.L.S.; Lana-Peixoto,M.A.; Teixeira,M.M.; Teixeira,A.L..
Multiple sclerosis (MS) is a chronic inflammatory demyelinating disease of the human central nervous system. Although its etiology is unknown, the accumulation and activation of mononuclear cells in the central nervous system are crucial to its pathogenesis. Chemokines have been proposed to play a major role in the recruitment and activation of leukocytes in inflammatory sites. They are divided into subfamilies on the basis of the location of conserved cysteine residues. We determined the levels of some CC and CXC chemokines in the cerebrospinal fluid (CSF) of 23 relapsing-remitting MS patients under interferon-ß-1a therapy and 16 control subjects using ELISA. MS patients were categorized as having active or stable disease. CXCL10 was significantly...

Expression of the chemokine receptor CXCR4 on lymphocytes of leprosy patients BJMBR
Mendonça,V.A.; Alvim de Melo,G.E.B.; Araújo,M.G.; Borges,V.O.; Costa,R.D.; Martins-Filho,O.A.; Teixeira- Carvalho,A.; Sathler-Avelar,R.; Teixeira,M.M.; Teixeira,A.L..
Leprosy is caused by Mycobacterium leprae, which induces chronic granulomatous infection of the skin and peripheral nerves. The disease ranges from the tuberculoid to the lepromatous forms, depending on the cellular immune response of the host. Chemokines are thought to be involved in the immunopathogenesis of leprosy, but few studies have investigated the expression of chemokine receptors on leukocytes of leprosy patients. In the present study, we evaluated 21 leprosy patients (M/F: 16/5) with a new diagnosis from the Dermatology Outpatient Clinic of the University Hospital, Federal University of Minas Gerais. The control group was composed of 20 healthy members (M/F: 15/5) of the community recruited by means of announcements. The expression of CCR2,...

Increased levels of glutamate in the central nervous system are associated with behavioral symptoms in experimental malaria BJMBR
Miranda,A.S.; Vieira,L.B.; Lacerda-Queiroz,N.; Souza,A.H.; Rodrigues,D.H.; Vilela,M.C.; Gomez,M.V.; Machado,F.S.; Rachid,M.A.; Teixeira,A.L..
Cerebral malaria (CM) is a severe complication resulting from Plasmodium falciparum infection. This condition has been associated with cognitive, behavioral and motor dysfunctions, seizures and coma. The underlying mechanisms of CM are incompletely understood. Glutamate and other metabolites such as lactate have been implicated in its pathogenesis. In the present study, we investigated the involvement of glutamate in the behavioral symptoms of CM. Seventeen female C57BL/6 mice (20-25 g) aged 6-8 weeks were infected with P. berghei ANKA by the intraperitoneal route using a standardized inoculation of 10(6) parasitized red blood cells suspended in 0.2 mL PBS. Control animals (N = 17) received the same volume of PBS. Behavioral and neurological symptoms were...

Serum levels of brain-derived neurotrophicfactor correlate with the number of T2 MRI lesions in multiple sclerosis BJMBR
Comini-Frota,E.R.; Rodrigues,D.H.; Miranda,E.C.; Brum,D.G.; Kaimen-Maciel,D.R.; Donadi,E.A.; Teixeira,A.L..
The objective of the present study was to determine if there is a relationship between serum levels of brain-derived neurotrophic factor (BDNF) and the number of T2/fluid-attenuated inversion recovery (T2/FLAIR) lesions in multiple sclerosis (MS). The use of magnetic resonance imaging (MRI) has revolutionized the study of MS. However, MRI has limitations and the use of other biomarkers such as BDNF may be useful for the clinical assessment and the study of the disease. Serum was obtained from 28 MS patients, 18-50 years old (median 38), 21 women, 0.5-10 years (median 5) of disease duration, EDSS 1-4 (median 1.5) and 28 healthy controls, 19-49 years old (median 33), 19 women. BDNF levels were measured by ELISA. T1, T2/FLAIR and gadolinium-enhanced lesions...

Use of inflammatory molecules to predict the occurrence of fever in onco-hematological patients with neutropenia BJMBR
Ribeiro,A.F. Tibúrcio; Nobre,V.; Neuenschwander,L.C.; Teixeira,A.L.; Xavier,S.G.; Paula,F.D.F.; Teixeira,M.M.; Teixeira,J.C.A.; Bittencourt,H..
Febrile neutropenia remains a frequent complication in onco-hematological patients, and changes in the circulating level of inflammatory molecules (IM) may precede the occurrence of fever. The present observational prospective study was carried out to evaluate the behavior of plasma tumor necrosis factor alpha (TNF-α), soluble TNF-α I and II receptors (sTNFRI and sTNFRII), monocyte chemoattractant protein-1 [MCP-1 or chemokine (c-c motif) ligand 2 (CCL2)], macrophage inflammatory protein-1α (MIP-1α or CCL3), eotaxin (CCL11), interleukin-8 (IL-8 or CXCL8), and interferon-inducible protein-10 (IP-10 or CXCL10) in 32 episodes of neutropenia in 26 onco-hematological patients. IM were tested on enrollment and 24-48 h before the onset of fever and within 24 h of...
